چکیده
ackground: Recent studies have raised concerns about subtle long-lasting neurobiological changes that might be triggered by xposure to Cannabis derivatives, especially in a critical phase of brain maturation, such as puberty. The mesolimbic dopamine (DA) ystem, involved in the processing of drug-induced reward, is a locus of action of cannabinoids and endocannabinoids. Thus, we ompared the effects of repeated cannabinoid administration in adolescent and adult rats on DA neuronal functions and responses o drugs of abuse. ethods: Single-unit extracellular recordings from antidromically identified mesoaccumbens DA neurons and from their target cells n the nucleus accumbens were carried out in urethane-anesthetized rats. Animals were pretreated during adolescence or adulthood, or 3 days, with the cannabinoid agonist WIN55212.2 (WIN) or vehicle and allowed a 2-week interval. esults: In cannabinoid-administered rats, DA neurons were significantly less responsive to the stimulating action of WIN, regardless f the age of pretreatment; however, in the adolescent group, but not in the adult, long-lasting cross-tolerance developed to morphine, ocaine, and amphetamine. onclusions: Our study suggests that an enduring form of neuronal adaptation occurs in DA neurons after subchronic cannabinoid ntake at a young age, affecting subsequent responses to drugs of abuse.

متن کاملCinnamaldehyde attenuates dopaminergic neuronal loss in substantia nigra and induces midbrain catalase activity in a mouse model of Parkinson’s disease
Background and Objective: Parkinson's disease (PD) is the second most common neurodegenerative disease after Alzheimer's disease that affects 3% of the population. PD involves a progressive degeneration of dopaminergic neurons in the substantia nigra pars compacta (SNc) and subsequent loss of dopamine. Dopamine depletion leads to movement dysfunction and is accompanied with tremor, rigid muscle...
متن کاملThe firing rate of neurons in the nucleus cuneiformis in response to formalin in male rat
Introduction: Although formalin-induced activity in primary afferent fibers and spinal dorsal horn is well described, the midbrain neural basis underlying each phase of behavior in formalin test has not been clarified. The present study was designed to investigate the nucleus cuneiformis (CnF) neuronal responses during two phases after subcutaneous injection of formalin into the hind paw...
متن کاملComparison of Rat Primary Midbrain Neurons Cultured in DMEM/F12 and Neurobasal Mediums
Introduction: Midbrain dopaminergic neurons are involved in various brain functions, including motor behavior, reinforcement, motivation, learning, and cognition. Primary dopaminergic neurons and also several lines of these cells are extensively used in cell culture studies. Primary dopaminergic neurons prepared from rodents have been cultured in both DMEM/F12 and neurobasal mediums in several ...
متن کاملRepeated administration of cannabinoid receptor agonist and antagonist impairs short and long term plasticity of rat’s dentate gyrus in vivo
Introduction: The effects of cannabinoids (CBs) on synaptic plasticity of hippocampal dentate gyrus neurons have been shown in numerous studies. However, the effect of repeated exposure to cannabinoids on hippocampal function is not fully understood. In this study, using field potential recording, we investigated the effect of repeated administration of the nonselective CB receptor agonist WIN5...
